metabolism | APPBP1 and UBA3 are homologous to the amino and carboxyl regions of the ubiquitin-activating enzyme, respectively. APPBP1-UBA3 first catalyzes the adenylation of C-terminal Gly76 of NEDD8 in the presence of Mg2+ and ATP and subsequently forms a thioester intermediate between its catalytic cysteine (Cys216 on UBA3) and the C terminus of NEDD8. The APPBP1-UBA3 complex ultimately binds the E2 of NEDD8, Ubc12, and promotes the transfer of NEDD8 onto the catalytic cysteine of Ubc12 | Homo sapiens |
